The peptide binds GHRH receptors on somatotroph cells within the anterior pituitary, triggering a signalling cascade that increases growth hormone secretion. Its improved resistance to dipeptidyl peptidase IV degradation distinguishes it from the parent hormone. In the DAC-bearing version, a maleimide group reacts with a cysteine residue on serum albumin, forming a covalent bond that keeps the peptide in circulation far longer. That albumin attachment is the central design feature separating the two research variants.
Pharmacokinetic behaviour differs sharply between the two forms. The DAC-bearing peptide shows an extended circulation time measured in days, whereas the version without the complex is cleared within roughly half an hour. This gap shapes how researchers design dosing schedules in animal models. Whether the prolonged presence of the DAC form produces effects meaningfully different from the short-acting variant remains an open question, since comparative human data are scarce.
Laboratory handling centers on minimizing exposure to water, heat and oxygen before use. Working solutions are typically prepared in sterile water or a mild buffer, and any residual particulate matter is removed by filtration. When the powder dissolves slowly, a small proportion of acetonitrile or dilute acetic acid is sometimes added as a co-solvent. Containers are kept sealed and desiccated between uses. Records of lot number, reconstitution date and storage conditions support later comparison of results across experiments.
Identity and purity are established with reversed phase high performance liquid chromatography coupled to mass spectrometry. The chromatographic step separates the target peptide from truncated sequences and deletion products, while the mass measurement confirms the expected molecular weight to within a fraction of a dalton. Because the two common variants differ by the presence of the linker, mass alone can distinguish them in the unconjugated state. Amino acid analysis and peptide mapping are used when sequence level confirmation is required.

Two forms circulate in research settings and are frequently confused. One carries the drug affinity complex and is often written as CJC-1295 with DAC; the other lacks that group and is usually called modified GRF(1-29). The two share the same core sequence but differ sharply in how long they persist in blood. Products labelled only as CJC-1295 normally refer to the version carrying the complex. Documentation that omits the distinction leaves the intended molecule ambiguous.
CJC-1295 is a synthetic peptide built as a long-acting analogue of growth hormone-releasing hormone. Its backbone matches the first twenty-nine residues of the natural human hormone, with four amino acid substitutions added to slow enzymatic breakdown. A reactive maleimide group, commonly termed the drug affinity complex, allows the peptide to attach to circulating albumin after administration. That albumin attachment keeps the molecule in the bloodstream for an extended period instead of being cleared within minutes.
Naming in this area is inconsistent, and readers should treat product labels with care. In much of the literature and in vendor catalogs, the unqualified term refers to the albumin-binding version, while the version lacking the DAC group appears as modified GRF(1-29), mod GRF(1-29), or the same name with a without-DAC qualifier. Because one abbreviation has been applied to both materials, the only reliable way to identify a sample is to check the stated sequence and the presence of the linker.
CJC-1295 is the name used for a synthetic peptide modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. The compound was described by a Canadian drug discovery company in the mid-2000s as a long-acting research tool. Two closely related molecules share the name in practice: one carries a drug affinity complex, or DAC, group, and one does not. The distinction matters because the two behave differently in circulation.
The peptide backbone corresponds to GRF(1-29), the first 29 residues of native growth hormone-releasing hormone, which retains most of the receptor-activating activity of the full-length molecule. Four substitutions distinguish the analog from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes slow cleavage by dipeptidyl peptidase IV, the enzyme that degrades native hormone in plasma within minutes. The outcome is improved enzymatic stability combined with a still brief residence time when no additional modification is present.
Analytical confirmation usually relies on reversed-phase high-performance liquid chromatography for purity and on liquid chromatography coupled to mass spectrometry for identity. Mass data reveal the expected molecular mass and can flag truncated or oxidized species. Amino acid analysis and peptide mapping provide sequence-level verification. Immunoassays are used in some biological matrices, but antibodies raised against one releasing-hormone analog may cross-react with another. Reported purity figures depend heavily on the method used, so comparisons between suppliers require matching the analytical approach.
The two variants differ dramatically in how long they persist in circulation. The form lacking the albumin-binding group has a plasma half-life measured in tens of minutes, comparable to the natural hormone fragment. The version carrying the drug affinity complex binds albumin and shows a half-life of roughly six to eight days in human studies. That figure comes from small trials that tracked hormone levels over extended periods. The physiological consequences of sustained versus pulsatile stimulation are still debated and the literature does not settle the point.
Lyophilized peptide powder is comparatively stable when kept dry, cold, and protected from light. Once dissolved, the molecule is vulnerable to deamidation, oxidation, and aggregation, with the rate depending on pH, buffer composition, and temperature. Alkaline conditions and repeated freeze-thaw cycles accelerate loss of the intact peptide. The methionine present in the native sequence is a known oxidation site, which is one reason it was replaced in the modified fragment. Suppliers typically recommend cold storage of solutions and use within a short window.
Although ribozymes are quite rare in most cells, their roles are sometimes essential to life. For example, the functional part of the ribosome, the biological machine that translates RNA into proteins, is fundamentally a ribozyme, composed of RNA tertiary structural motifs that are often coordinated to metal ions such as Mg2+ as cofactors. In a model system, there is no requirement for divalent cations in a five-nucleotide RNA catalyzing trans-phenylalanation of a four-nucleotide substrate with 3 base pairs complementary with the catalyst, where the catalyst/substrate were devised by truncation of the C3 ribozyme. The best-studied ribozymes are probably those that cut themselves or other RNAs, as in the original discovery by Cech and Altman. However, ribozymes can be designed to catalyze a range of reactions or they have them naturally. Some of these activities include the following:
== Function == Serpin A12 is a protease inhibitor with an approximate weight of 47 kDa and is a member of the adipokine family of cytokines excreted by adipose tissue. Members of this family regulate a number of cellular processes, such as inflammation mediation and insulin resistance. Made up of 414 amino acids, its main function is modulating the insulin inhibiting protease KLK7, mainly in adipose tissue. Among other functions, Serpin A12 performs insulin-sensitizing actions. Serpin A12 treatment of obese and insulin-resistant mice has been shown to decrease the expression of insulin resistance genes in white adipose tissue as well as improving carbohydrate resistance. Serpin A12 also increases bone density, which helps prevent osteoporosis. It does so by regulating osteoblasts, assisting in their mineralization of the bone matrix, thus balancing bone formation with bone resorption.
== Detergents, salts & enzymes == Cell lysis is a critical step in the purification of enzymes from bacterial cells, various components are commonly included in lysing buffers to facilitate effective cell disruption and release of the target enzyme. These components include detergents, salts, and enzymes, each playing a specific role in the lysis process. Examples of detergents used in lysing buffers include: Detergents: Detergents are amphipathic molecules that possess both hydrophilic and hydrophobic properties. In the context of cell lysis, detergents act by disrupting the lipid bilayer of the bacterial cell membrane, leading to membrane permeabilization and release of intracellular components, including the target enzyme. Commonly used detergents in lysing buffers include: a. Triton X-100: a nonionic detergent frequently employed due to its mild and effective membrane-disrupting properties, it solubilizes lipids and membrane proteins, allowing the release of intracellular contents. b. Sodium dodecyl sulfate (SDS): an anionic detergent that denatures proteins by disrupting their secondary and tertiary structures, it solubilizes cellular membranes and aids in protein extraction. c. Tween-20: a nonionic detergent is milder compared to SDS and Triton X-100. It assists in membrane permeabilization and solubilization of proteins without causing significant denaturation. Salts: Salts are crucial components of lysing buffers as they help maintain optimal cellular conditions and provide ionic strength to facilitate cell disruption.
De Duve's work has contributed to the emerging consensus towards accepting the endosymbiotic theory; which idea proposes that organelles in eukaryotic cells originated as certain prokaryotic cells that came to live inside eukaryotic cells as endosymbionts. According to de Duve's version, eukaryotic cells with their structures and properties, including their ability to capture food by endocytosis and digest it intracellularly, developed first. Later, prokaryotic cells were incorporated to form more organelles. De Duve proposed that peroxisomes, which allowed cells to withstand the growing amounts of free molecular oxygen in the early-Earth atmosphere, may have been the first endosymbionts. Because peroxisomes have no DNA of their own, this proposal has much less evidence than similar claims for mitochondria and chloroplasts. His later years were mostly devoted to origin of life studies, which he admitted was still a speculative field (see thioester).
The possibility of a militarized Japan has also been a continuous concern to the Chinese leadership since the late 1990s. In addition, China's military leadership has been reacting to and learning from the successes and failures of the United States Armed Forces during the Kosovo War, the 2001 invasion of Afghanistan, the 2003 invasion of Iraq, and the Iraqi insurgency. All these lessons inspired China to transform the PLA from a military based on quantity to one based on quality. Chairman Jiang Zemin officially made a "revolution in military affairs" (RMA) part of the official national military strategy in 1993 to modernize the Chinese armed forces. A goal of the RMA is to transform the PLA into a force capable of winning what it calls "local wars under high-tech conditions" rather than a massive, numbers-dominated ground-type war. Chinese military planners call for short decisive campaigns, limited in both their geographic scope and their political goals. In contrast to the past, more attention is given to reconnaissance, mobility, and deep reach. This new vision has shifted resources towards the navy and air force. The PLA is also actively preparing for space warfare and cyber-warfare. In 2002, the PLA began holding military exercises with militaries from other countries. From 2018 to 2023, more than half of these exercises have focused on military training other than war, generally antipiracy, or antiterrorism exercises involving combatting non-state actors.

Liver chestnut or dark chestnut are not a separate genetic color, but a descriptive term. The genetic controls for the depth of shade are not presently understood. Liver chestnuts are a very dark-reddish brown. Liver chestnuts are included in the term "dark chestnut." The darkest chestnuts, particularly common in the Morgan horse, may be indistinguishable from true black without very careful inspection. Often confusingly called "black chestnuts", they may be identified by small amounts of reddish hair on the lower legs, mane and tail, or by DNA or pedigree testing. Recently, it has been suggested that the trait or traits that produce certain darker shades of chestnut and bay, referred to as "sooty" coloration follow a recessive mode of inheritance.
Veneer is obtained either by "peeling" the trunk of a tree or by slicing large rectangular blocks of wood known as flitches. The appearance of the grain and figure in wood comes from slicing through the growth rings of a tree and depends upon the angle at which the wood is sliced. There are three main types of veneer-making equipment used commercially:
=== Axon growth regulation === Crosslinking anti-Thy-1 Ab can promote neurite outgrowth which is dependent on G{alpha}i and L- and N-type calcium channel activation. The ligand for promotion of neurite outgrowth on astrocytes is not yet identified, but the inhibitory ligand has been suggested to be integrins. Thy1 is one of the known ligands of beta 3 integrins. Interaction of thy1 expressed on maturing axons with beta 3 integrins expressed on mature astrocytes is one of the causes of halting of axon growth.
I never take any psychedelic, have a psychedelic experience, without having that feeling of, "I don't know what's going to happen." In that sense, it's still fundamentally an enigma and a mystery." Bill Gates implied in an interview with Playboy that he tried LSD during his youth. Aldous Huxley, author of Brave New World, became a user of psychedelics after moving to Hollywood. He was at the forefront of the counterculture's use of psychedelic drugs, which led to his 1954 work The Doors of Perception. Dying from cancer, he asked his wife on 22 November 1963 to inject him with 100 μg of LSD. He died later that day. Steve Jobs, co-founder and former CEO of Apple Inc., said, "Taking LSD was a profound experience, one of the most important things in my life." Ernst Jünger, German writer and philosopher, throughout his life had experimented with drugs such as ether, cocaine, and hashish; and later in life he used mescaline and LSD. These experiments were recorded comprehensively in Annäherungen (1970, Approaches). The novel Besuch auf Godenholm (1952, Visit to Godenholm) is clearly influenced by his early experiments with mescaline and LSD. He met with LSD inventor Albert Hofmann and they took LSD together several times. Hofmann's memoir LSD, My Problem Child describes some of these meetings. In a 2004 interview, Paul McCartney said that The Beatles' songs "Day Tripper" and "Lucy in the Sky with Diamonds" were inspired by LSD trips.
=== Chemoenzymatic labeling === Chemoenzymatic labeling provides an alternative strategy to incorporate handles for click chemistry. The Click-IT O-GlcNAc Enzymatic Labeling System, developed by the Hsieh-Wilson group and subsequently commercialized by Invitrogen, utilizes a mutant GalT Y289L enzyme that is able to transfer azidogalactose (GalNAz) onto O-GlcNAc. The presence of GalNAz (and therefore also O-GlcNAc) can be detected with various alkyne-containing probes with identifiable tags such as biotin, dye molecules, and PEG.

Tool use by animals may indicate different levels of learning and cognition. The sea otter uses rocks as essential and regular parts of its foraging behavior (smashing abalone from rocks or breaking open shells), with some populations spending 21% of their time making tools. Other tool use, such as chimpanzees using twigs to "fish" for termites, may be developed by watching others use tools and may even be a true example of animal teaching. Tools may even be used in solving puzzles in which the animal appears to experience a "Eureka moment". Other mammals that do not use tools, such as dogs, can also experience a Eureka moment. Brain size was previously considered a major indicator of the intelligence of an animal. Since most of the brain is used for maintaining bodily functions, greater ratios of brain to body mass may increase the amount of brain mass available for more complex cognitive tasks. Allometric analysis indicates that mammalian brain size scales at approximately the 2⁄3 or 3⁄4 exponent of the body mass. Comparison of a particular animal's brain size with the expected brain size based on such allometric analysis provides an encephalization quotient that can be used as another indication of animal intelligence. Sperm whales have the largest brain mass of any animal on earth, averaging 8,000 cubic centimetres (490 cu in) and 7.8 kilograms (17 lb) in mature males. Self-awareness appears to be a sign of abstract thinking. Self-awareness, although not well-defined, is believed to be a precursor to more advanced processes such as metacognitive reasoning.

== The MEROPS M9 family == This group of metallopeptidases constitutes the MEROPS peptidase family M9, subfamilies M9A and M9B (microbial collagenase, clan MA(E)). The protein fold of the peptidase domain for members of this family resembles that of thermolysin, the type example for clan MA and the predicted active site residues for members of this family and thermolysin occur in the motif HEXXH. Microbial collagenases have been identified from bacteria of both the Vibrio and Clostridium genera. Collagenase is used during bacterial attack to degrade the collagen barrier of the host during invasion. Vibrio bacteria are sometimes used in hospitals to remove dead tissue from burns and ulcers. Clostridium histolyticum is a pathogen that causes gas gangrene; nevertheless, the isolated collagenase has been used to treat bed sores. Collagen cleavage occurs at an Xaa+Got in Vibrio bacteria and at Yaa+Gly bonds in Clostridium collagenases. Analysis of the primary structure of the gene product from Clostridium perfringens has revealed that the enzyme is produced with a stretch of 86 residues that contain a putative signal sequence. Within this stretch is found PLGP, an amino acid sequence typical of collagenase substrates. This sequence may thus be implicated in self-processing of the collagenase. Metalloproteases are the most diverse of the seven main types of protease, with more than 50 families identified to date. In these enzymes, a divalent cation, usually zinc, activates the water molecule. The metal ion is held in place by amino acid ligands, usually three in number.
Many types of breeder reactor are possible: A "breeder" is simply a nuclear reactor designed for very high neutron economy with an associated conversion ratio higher than 1.0. In principle, almost any reactor design could be tweaked to become a breeder. For example, the reduced-moderation water reactor concept uses mixed-oxide fuel in a tight lattice to increase the conversion ratio. Aside from water-cooled, there are many other types of breeder reactor currently envisioned as possible. These include molten-salt cooled, gas cooled, and liquid-metal cooled designs in many variations. Almost any of these basic design types may be fueled by uranium, plutonium, many minor actinides, or thorium, and they may be designed for many different goals, such as creating more fissile fuel, long-term steady-state operation, or active burning of nuclear wastes. Extant reactor designs are sometimes divided into two broad categories based upon their neutron spectrum, which generally separates those designed to use primarily uranium and transuranics from those designed to use thorium and avoid transuranics. These designs are:
== Composition == Tecemotide is a synthetic lipopeptide that is 27 amino acids long. Its molecular formula is C124H203N33O38, and its amino acid sequence is S T A PPAH G VTSAPDTRPAPGSTAPPKG. The first 25 amino acids of tecemotide are derived from the mucin 1 (MUC1, carcinoma-associated mucin, episialin, or CD227) sequence. The 26th modified amino acid, K, is palmityl-lysine (N6-(1-oxohexadecyl)-L-lysine), and the 27th is glycine.

The synthetic peptide keeps the receptor-binding region of GHRH but carries substitutions that resist enzymatic cleavage. In the DAC version, an added group also anchors the molecule to albumin, extending its presence in the bloodstream.
The maleimide group forms a covalent link with albumin, an abundant blood protein. This attachment slows clearance, so a single administration persists much longer than the unmodified peptide. The feature is the main reason the two variants are handled differently in study design.
It is not an approved therapeutic product in major regulatory jurisdictions. It appears in research chemical catalogs and scientific literature rather than pharmacy shelves. Clinical development was limited and did not reach approval.
The mass difference from the linker is large enough for routine detection by mass spectrometry. The unconjugated form gives a single sharp signal at its expected weight. Material that has already reacted with albumin shows a much higher mass and a broadened chromatographic peak.
