The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
Laboratory handling centers on minimizing exposure to water, heat and oxygen before use. Working solutions are typically prepared in sterile water or a mild buffer, and any residual particulate matter is removed by filtration. When the powder dissolves slowly, a small proportion of acetonitrile or dilute acetic acid is sometimes added as a co-solvent. Containers are kept sealed and desiccated between uses. Records of lot number, reconstitution date and storage conditions support later comparison of results across experiments.
Identity and purity are established with reversed phase high performance liquid chromatography coupled to mass spectrometry. The chromatographic step separates the target peptide from truncated sequences and deletion products, while the mass measurement confirms the expected molecular weight to within a fraction of a dalton. Because the two common variants differ by the presence of the linker, mass alone can distinguish them in the unconjugated state. Amino acid analysis and peptide mapping are used when sequence level confirmation is required.
Lyophilized material is generally stable for extended periods when held at minus twenty degrees Celsius or below and protected from moisture and light. In solution the peptide is more labile; bond hydrolysis, aggregation and oxidation of susceptible residues all proceed faster at ambient temperature. Repeated freeze and thaw cycles should be avoided because they promote clumping and loss of soluble material. The conjugated variant adds a further consideration, since the maleimide group can hydrolyze in aqueous buffer and lose its ability to react with albumin.

Clearance profiles diverge sharply between the two versions. The albumin-binding molecule stays in plasma for several days, whereas the unmodified analog is largely gone within about half an hour in reported work. Cleavage by dipeptidyl peptidase IV is a major contributor to the short life of the unmodified sequence. These gaps mean the two versions cannot be substituted for each other in study design or in reading results side by side.
Reports on this compound commonly follow serum growth hormone and insulin-like growth factor 1 across defined time windows. Protocols differ in sampling frequency, assay platform, and participant characteristics, which makes direct comparison between publications difficult. Some work focuses on pulsatile release patterns instead of average concentrations. Whether repeated exposure alters endogenous hormone rhythms over long periods remains an open question, and the formal literature is thinner than the volume of informal commentary implies.
The core sequence keeps the receptor-binding region of GHRH while replacing four positions that are vulnerable to dipeptidyl peptidase-4 and other proteases. Substitutions at positions 2, 8, 15, and 27 raise metabolic stability relative to the natural hormone. The N-terminal residues remain essential for activity, so changes there generally lower potency. Molecular weight sits near 3368 daltons for the tetrasubstituted analog without the linker, while the albumin-binding form is heavier because of the added maleimide group.
CJC-1295 is a synthetic peptide modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. Its sequence corresponds to the first twenty-nine residues of human GHRH, with four substitutions that slow enzymatic breakdown. Early descriptions placed the compound in research on growth hormone deficiency and related conditions, and later literature groups it with the long-acting GHRH analogs. The name appears in both laboratory and popular fitness writing, where it sometimes labels chemically different peptides.
== Involvement in ProteomeXchange == The ProteomeXchange consortium has been set up to provide a coordinated submission of MS proteomics data to the main existing proteomics repositories, and to encourage optimal data dissemination. The consortium contains several member databases, including PRIDE and PeptideAtlas. The earliest conception of ProteomeXchange stems from a meeting at the HUPO 2005 conference in Munich, where the main proteomics data repositories at the time agreed in principle to exchange their data, and thus provide a means for the user to find public proteomics data at any of the participating databases. Due to the rapid development of the field, and the need to first develop suitable standards for data exchange, it took almost ten years from that meeting to actually implement this system, an effort that was funded by the 'ProteomeXchange' Coordination Action grant of the European Commission's Seventh Framework Programme.
In October 2025, Russian Foreign Ministry spokeswoman Maria Zakharova said there was no cold war with NATO because it had already developed into a "firey" conflict, and accused NATO of lying about Russian sabotage to justify vast military spending.
=== Czech Republic === After joining NATO, the Army of the Czech Republic developed a combat ration known as the BDP (Czech: Bojová Dávka Potravin). The BDP comes in two versions, type I and II, each holding two ready-to-eat main courses packed in large foil "cans" (beef roast with rice, pork goulash with potato, spicy risotto, pork with carrots and vegetables, etc.), a small plastic cup of lunch meat spread, cheese spread, hard bread, cookies, jam, instant coffee, tea bags, fruit-flavored multivitamin drink tablets, vitamin C enriched fruit drink powder, a chocolate bar, sugar, salt, chewing gum, wet napkins, paper towels, a plastic bag, and a menu and instruction sheet. A modified version of the BDP known as the KDP (Czech: Konzervovaná Dávka Potravin) is also used. This contains the same items as the BDP, but adds an aluminium cup, plastic utensils, a folding stove with fuel tablets and matches, and soap.
=== February–April 1881 === On 14 February Carrington captured Ramokoatsi, which overlooked the main road to Morija. The following day, a force of 370 cavalry, 100 infantry, 50 native levies, and three artillery pieces under Brabant was sent out in search of a new camping ground. Upon crossing a spruit in the Ramibidikwa area, a CMR scout reported a massed formation of Basuto horsemen. Brabant ordered his soldiers to form a square; soon afterwards the Basuto commenced an attack on its front and two flanks. A combination of rifle and case shot fire kept the Basuto at bay in the center and the right flank. On the left, the Basuto managed to almost reach melee range before being likewise driven off. The artillery continued to fire on the retreating Basuto, who suffered 138 casualties in the engagement. One month later Clarke assumed personal command of the force, moving the camp to Ramibidikwa, 20 miles (32 km) from Morija. On 22 March, Carrington was heavily wounded in the vicinity of the new camp. By early April, Sprigg's conduct of the war was being heavily criticized in the Cape Parliament, whose opposition members were pushing for a vote of no confidence. Using Letsie as an intermediary, Robinson organized a meeting between Griffith and Lerotholi outside Maseru on 17 April. The two sides signed an armistice, although Lerotholi was unwilling to surrender his weapons, as the motion would be too unpopular among his tribesmen. On 29 April, Robinson announced the peace settlement, known as the Award.
A few examples of character strength according to Character Strength and Virtues: A Handbook and Classification by Seligman and Peterson (2004) are bravery, integrity, citizenship, humility, prudence, gratitude, and hope, all of which are shown in the rise to recovery. Thirdly, the Meaningful Life is service and membership to positive organizations. Examples of positive organizations include family, workplace, social groups, and society in general. Membership of these groups fosters positive affect, while also promoting character strengths, which as seen in the Engaged Life, can aid in beating addiction.

The binding of tyrosine hydroxylase to membranes involves the N-terminal region of the enzyme, and may be regulated by a three-way interaction between 14-3-3 proteins, the N-terminal region of tyrosine hydroxylase, and negatively charged membranes. Tyrosine hydroxylase can also be regulated by inhibition. Phosphorylation at Ser40 relieves feedback inhibition by the catecholamines dopamine, epinephrine, and norepinephrine. The catecholamines trap the active-site iron in the Fe(III) state, inhibiting the enzyme. It has been shown that the expression of tyrosine hydroxylase can be affected by the expression of SRY. The down regulation of the SRY gene in the substantia nigra can result in a decrease in tyrosine hydroxylase expression. Long term regulation of tyrosine hydroxylase can also be mediated by phosphorylation mechanisms. Hormones (e.g. glucocorticoids), drugs (e.g. cocaine), or second messengers such as cAMP increase tyrosine hydroxylase transcription. Increase in tyrosine hydroxylase activity due to phosphorylation can be sustained by nicotine for up to 48 hours. Tyrosine hydroxylase activity is regulated chronically (days) by protein synthesis.
April 3 – A United States Air Force F-15E Strike Eagle is shot down over Iran during combat operations. Both crew members are later rescued. During the rescue mission, an A-10 Thunderbolt II is also hit and crashes, though its pilot ejects safely and is recovered. April 4 – 2025–26 NHL season: In ice hockey, the Buffalo Sabres clinched the Stanley Cup playoffs for the first time since the 2010–11 season, ending their 14-year Stanley Cup playoff drought. The average age of participants was 66 years and more participants were male (77%) than female. To determine the drug's effectiveness, investigators examined the occurrence of cardiovascular death, hospitalization for heart failure, and urgent heart failure visits. Participants were randomly assigned to receive a once-daily dose of either 10 mg of dapagliflozin or a placebo (inactive treatment). After about 18 months, people who received dapagliflozin had fewer cardiovascular deaths, hospitalizations for heart failure, and urgent heart-failure visits than those receiving the placebo. In July 2020, the FDA granted AstraZeneca a Fast Track Designation in the US for the development of dapagliflozin to reduce the risk of hospitalization for heart failure or cardiovascular death in adults following a heart attack. In August 2020, detailed results from the Phase III DAPA-CKD trial reportedly showed that dapagliflozin on top of standard of care reduced the composite measure of worsening of renal function or risk of cardiovascular or renal death by 39% compared to placebo (p<0.0001) in patients with chronic kidney disease stages 2–4 and elevated urinary albumin excretion. The results were consistent in patients both with and without type 2 diabetes. In April 2021, the FDA expanded the indications for dapagliflozin to include reducing the risk of kidney function decline, kidney failure, cardiovascular death, and hospitalization for heart failure in adults with chronic kidney disease who are at risk of disease progression.

The two share the same 29-amino-acid backbone. CJC-1295 carries an additional albumin-binding moiety that markedly extends its residence time in circulation, while the form without that moiety clears faster. Literature sometimes applies the name loosely to either variant.
It has not been approved by major regulatory agencies for any therapeutic indication. Material sold under this name is typically distributed for laboratory research only. Clinical use is not supported by large controlled trials.
Early human studies of the albumin-binding form reported circulation half-lives on the order of several days. The value depends on assay method and study design. Estimates for the form without the binding moiety are considerably shorter.
The mass difference from the linker is large enough for routine detection by mass spectrometry. The unconjugated form gives a single sharp signal at its expected weight. Material that has already reacted with albumin shows a much higher mass and a broadened chromatographic peak.
