CJC-1295 belongs to a class of synthetic peptides modeled on growth hormone releasing hormone, a forty-four amino acid signal produced by the hypothalamus. The compound is built from the first twenty-nine residues of the natural sequence, with four substitutions introduced at positions 2, 8, 15 and 27. These changes were designed to slow enzymatic breakdown while preserving receptor activation. The result is a peptide that is shorter than native GHRH and considerably more resistant to ordinary clearance pathways in circulation.
Two related forms appear under the CJC-1295 name. The simpler analogue, commonly written as modified GRF(1-29), carries the four substitutions but no additional conjugation. The second form attaches a maleimidopropionic acid linker to a lysine residue, a modification frequently called the drug affinity complex. That linker reacts with the free thiol on serum albumin to form a covalent bond. Because albumin has a long residence time in blood, the conjugated peptide stays in circulation far longer than the unconjugated analogue.
Receptor level activity follows the canonical GHRH pathway. The peptide binds the GHRH receptor, a class B G protein coupled receptor on somatotroph cells of the anterior pituitary. Binding raises intracellular cyclic AMP, which promotes calcium entry and the release of growth hormone into the bloodstream. Because the molecule acts at the same receptor as the endogenous hormone, its effect is superimposed on the natural pulsatile rhythm rather than replacing it. Whether sustained receptor occupation leads to desensitization is not fully settled.
Research material is normally supplied as a freeze-dried powder in sealed vials. In that state the peptide is comparatively robust, but prolonged exposure to warmth, moisture, or light accelerates degradation. Storage at minus twenty degrees Celsius or lower, with desiccant and protection from light, is the commonly described practice. Vials should be allowed to reach room temperature before opening to limit condensation on the powder. Moisture uptake during handling is a recognized source of variability in later measurements.
Reconstitution is typically performed with sterile water or bacteriostatic water, added slowly against the vial wall. The resulting solution should be clear and colorless; cloudiness or visible particles suggest a problem with the material or the diluent. Once in solution, the peptide is less stable than the dry powder. Refrigerated storage at two to eight degrees Celsius is common for short-term holding, while freezing aliquots is described for longer periods.
Purity is most often assessed by reversed-phase high-performance liquid chromatography, reported as a percentage of total peak area. Identity is confirmed by mass spectrometry, which yields a molecular ion consistent with the expected sequence. Amino acid analysis and peptide mapping provide additional characterization. Reported purity values are method-dependent, so figures from different laboratories are not always directly comparable without details of column, gradient, and detection wavelength.

Both forms act at the pituitary receptor for growth hormone-releasing hormone and increase growth hormone output, which in turn raises insulin-like growth factor 1. A long-acting analog produces sustained rather than pulsatile stimulation, and the physiological consequences of that pattern are not fully settled. Published human data on the extended form remain limited, and much of what circulates in discussion traces to early company reports rather than independent replication. How sustained exposure affects normal feedback remains an open question.
The distinguishing feature of the DAC form is a maleimide-containing group that reacts with the free thiol of cysteine-34 on human serum albumin. This reaction forms a covalent bond without enzymatic assistance, and it takes place after the peptide enters the bloodstream. Because albumin is abundant and long-lived, the attached peptide is carried through circulation far longer than an unmodified fragment would survive. The chemistry is a deliberate pharmacokinetic strategy rather than a change to receptor activity.
Enzymatic protection is a separate mechanism from plasma protein binding. The four substitutions in the backbone reduce recognition by dipeptidyl peptidase IV, which normally cleaves the natural hormone within minutes. Without the reactive group, this resistance still yields only a short window of activity, generally reported in the range of tens of minutes. With it, reported half-lives in early human work extended to several days. The size of that gap is the main practical distinction between the two materials.
Two related peptides circulate under the CJC-1295 label, and they differ mainly in how long they persist in circulation. The version carrying a drug affinity complex includes a maleimidopropionic acid linker that forms a covalent bond with serum albumin. The other version, usually written as modified GRF(1-29) or tetrasubstituted GRF(1-29), lacks that linker and is cleared quickly. Mixing the two produces inconsistent readings of published half-life values, because the linker rather than the receptor-facing sequence drives most of the difference.
The core sequence keeps the receptor-binding region of GHRH while replacing four positions that are vulnerable to dipeptidyl peptidase-4 and other proteases. Substitutions at positions 2, 8, 15, and 27 raise metabolic stability relative to the natural hormone. The N-terminal residues remain essential for activity, so changes there generally lower potency. Molecular weight sits near 3368 daltons for the tetrasubstituted analog without the linker, while the albumin-binding form is heavier because of the added maleimide group.
SWAPO leaders soon went abroad to mobilise support for their goals within the international community and newly independent African states in particular. The movement scored a major diplomatic success when it was recognised by Tanganyika and allowed to open an office in Dar es Salaam. SWAPO's first manifesto, released in July 1960, was remarkably similar to SWANU's. Both advocated the abolition of colonialism and all forms of racialism, the promotion of Pan-Africanism, and called for the "economic, social, and cultural advancement" of South West Africans. However, SWAPO went a step further by demanding immediate independence under black majority rule, to be granted at a date no later than 1963. The SWAPO manifesto also promised universal suffrage, sweeping welfare programmes, free healthcare, free public education, the nationalisation of all major industry, and the forcible redistribution of foreign-owned land "in accordance with African communal ownership principles". Compared to SWANU, SWAPO's potential for wielding political influence within South West Africa was limited, and it was accordingly likelier to accept armed insurrection as the primary means of achieving its goals. SWAPO leaders also argued that a decision to take up arms against the South Africans would demonstrate their superior commitment to the nationalist cause. They believed that this would distinguish SWAPO from SWANU in the eyes of international supporters as the genuine vanguard of the Namibian independence struggle, and the legitimate recipient of any material assistance that was forthcoming.
methylation The covalent attachment of a methyl group (–CH3) to a chemical compound, protein, or other biomolecule, either spontaneously or by enzymatic catalysis. Methylation is one of the most widespread natural mechanisms by which nucleic acids and proteins are labelled. The methylation of nucleobases in a DNA molecule inhibits recognition of the methylated sequence by DNA-binding proteins, which can effectively silence the expression of genes. Specific residues within histones are also commonly methylated, which can change nucleosome positioning and similarly activate or repress nearby loci. The opposite reaction is demethylation.
The country had a 2019 Forest Landscape Integrity Index mean score of 5.95/10, ranking it 90th globally out of 172 countries. Some 3,700 plant species have been identified in the country, from which to date 23 have been declared natural monuments, 74 extinct, 39 endangered, 171 vulnerable, and 1,253 rare. The fauna of Romania consists of 33,792 species of animals, 33,085 invertebrate and 707 vertebrate, with almost 400 unique species of mammals, birds, reptiles, and amphibians, including about 50% of Europe's (excluding Russia) brown bears and 20% of its wolves. Romania is often seen by most as having the shape resembling one of a goldfish.
=== Differential diagnosis === About 99% of HD diagnoses based on the typical symptoms and a family history of the disease are confirmed by genetic testing to have the expanded trinucleotide repeat that causes HD. Most of the remaining are called HD-like (HDL) syndromes. The cause of most HDL diseases is unknown, but those with known causes are due to mutations in the prion protein gene (HDL1), the junctophilin 3 gene (HDL2), a recessively inherited unknown gene (HDL3—only found in two families and poorly understood), and the gene encoding the TATA box-binding protein (SCA17, sometimes called HDL4). Other autosomal dominant diseases that can be misdiagnosed as HD are dentatorubral-pallidoluysian atrophy and neuroferritinopathy. Also, some autosomal recessive disorders resemble sporadic cases of HD. These include chorea acanthocytosis and pantothenate kinase-associated neurodegeneration. One X-linked disorder of this type is McLeod syndrome.

== Adverse effects == The most common reported adverse effects were nausea (19%), headache (20%), and implant site reactions (21%). Uncommon but potentially serious hypersensitivity reactions including anaphylaxis were also reported. Afamelanotide was also found to sometimes induce darkening of certain pre-existing skin conditions, so regular full body skin examinations every 6 months is often recommended. Other common adverse effects reported during the clinical trials include back pain, upper respiratory tract infections, melanocyte naevus, decreased appetite, migraine, dizziness, weakness, fatigue, lethargy, sleepiness, hot flashes, abdominal pain, diarrhea, vomiting, flushing, development of warts, spots, and freckles, and itchy skin (between 1% and 10% of people). Uncommon and rare adverse effects include cystitis, folliculitis, gastrointestinal infections, hypersensitivity reactions, changes in appetite, depression, insomnia, balance disorders, lethargy, restless leg syndrome, syncope, photophobia, presbyopia, tinnitus, confusion, palpitations, hypertension, hypercholesterolaemia, and weight gain.
kinetochore A disc-shaped protein complex which assembles around the centromere of a chromosome during prometaphase of mitosis and meiosis, where it functions as the attachment point for microtubules of the spindle apparatus.
=== Christmas bombings === On 14 December 1972, Nixon sent an ultimatum demanding that Tho return to Paris to "negotiate seriously" within 72 hours or else he would bomb North Vietnam without limit. Knowing that Nixon was considering sacking him, Kissinger approved of his decision to resume bombing North Vietnam. Kissinger told the media that while the peace agreement was "99 percent completed…we will not be blackmailed into an agreement. We will not be stampeded into an agreement and, if I may say so, we will not be charmed into an agreement until its conditions are right". At the same time, Nixon ordered Admiral Thomas Hinman Moorer, the Chairman of the Joint Chiefs of Staff: "I don't want any more of this crap about the fact that we couldn't hit this target or that one. This is your chance to use military power to win this war, and if you don't, I'll hold you responsible". Following the rejection of Nixon's ultimatum, on 18 December, Operation Linebacker II was launched, the so-called Christmas Bombings that lasted until 29 December 1972. During these 11 days of bombing that were the heaviest bombing of the entire war, B-52 bombers flew 3,000 sorties and dropped 40,000 tons of bombs on Hanoi and Haiphong. About 1,261 people were killed in Hanoi and another 305 in Haiphong, as the North Vietnamese authorities had pulled out most people from the two cities beforehand to escape the expected bombings.
=== Medical === Puffballs are a known styptic and have long been used as wound dressing, either in powdered form or as slices 3 cm thick. Authors Hui-Yeng Y. Yap, Mohammad Farhan Ariffeen Rosli, et al. found evidence to suggest that C. gigantea was "traditionally used by American Indians, Nigerian and German folks" for this purpose. The authors, however, did not specify the preferred form of wound dressing (e.g., powdered or sliced). In addition to consuming the mushroom, the Māori people of New Zealand used it to stem bleeding and treat burns.
Since 1953, six Nobel prizes have been awarded in the area of polymer science, excluding those for research on biological macromolecules. This further testifies to its impact on modern science and technology. As Lord Todd summarised in 1980, "I am inclined to think that the development of polymerization is perhaps the biggest thing that chemistry has done, where it has had the biggest effect on everyday life".

The names are often used interchangeably in casual writing, but they are not strictly identical. Modified GRF(1-29) refers to the unconjugated analogue carrying only the four substitutions. The version sold as CJC-1295 with DAC includes the albumin binding linker and clears much more slowly.
DAC stands for drug affinity complex, a term describing the maleimidopropionic acid linker. This group forms a covalent bond with the cysteine residue on serum albumin after administration. The resulting adduct is what extends the circulating half life from minutes to days.
No. It is entirely synthetic and does not correspond to any known endogenous peptide. The design starts from human growth hormone releasing hormone and introduces deliberate substitutions. Any biological effect comes from mimicking the natural hormone at its receptor.
Cool, dark, and dry conditions are standard, with storage at minus twenty degrees Celsius or below. Desiccant and sealed vials limit moisture uptake. Repeated warming and cooling of the container is generally avoided.
